Protein translation in eukaryotes involves a complex interaction of multiple initiation and elongation factors that bind to mRNA to facilitate protein synthesis. 4E-Binding Protein 1 (4E-BP1) is a 12.4kDa protein that prevents initiation of mRNA translation by binding to eukaryotic initiation factor (eIF) 4E, thereby blocking its association with the 5’ cap structure present on mRNA. Phosphorylation of 4E-BP1 at several key residues reduces its affinity for eIF4E. 4E-BP1 is an effector of mTOR signalling, which plays a key role in obesity and insulin resistance. The 4E-BP1 Total Human Assay Kit provides a highly specific and convenient method to detect both phosphorylated and non-phosphorylated forms of 4E-BP1. Measurement of total 4E-BP1 protein offers the ability to normalize levels of phosphorylated 4E-BP1 under various cellular conditions as can be accomplished using the 4E-BP1 (human) (phospho-Thr37/46) EIA Kit (Prod. No. CAY-10011222).
